You can charter a small plane to take you to the airport in Las Terrenas/El Portillo. There are no lights at the airfield, so you need to land there during the day.
From Santo Domingo, you can take a Caribe Tours or Metro bus to Sanchez, the town just before getting to Samana. Vehicles meet the bus in Sanchez to take people to Las Terrenas.
Car rental also an option. But if you don't want a car all the time, I'd recommend one of the other options. Then renting a car in Las Terrenas for the days you want to explore by car.
