Shouldn't it be "Donde esta la casa de Pepe?"
or is it "Donde es???"

Use verb estar to talk about a location:
¿Dónde está la casa? Está en Santiago.

Use verb ser to talk about an event:
¿Dónde es la reunión? (Where is the meeting?)
